Ana-Maria LENNON-DUMENIL (Institut Curie – U932 INSERM , Paris – FRANCE): « Dentritic cell migration : from microfluidics to in vivo imaging »
Immature dendritic cells (DCs) patrol the interstitial space of peripheral tissues thanks to their remarkable antigen macropinocytic and phagocytic capacity. Encounter with danger-associated antigens triggers a developmental program referred to as “DC maturation”, which modifies many of their cell biological properties. This includes the up-regulation of the chemokine receptor CCR7, which allows mature DCs entering into lymphatic vessels and migrate to lymph nodes to activate T lymphocytes, as well as the down-regulation of macropinocytosis. In my talk, I will show how regulation of actin nucleation by Arp2/3 and Formins differentially regulates migration and macropinocytosis in immature and mature DCs (Vargas et al. Nat.Cell Biol. 2016) and discuss unpublished results highlighting that macropinocytosis (1) links nutrient sensing to DC migration and (2) impacts on the ability of these cells to move in environments displaying high hydraulic resistance.
