Pr. Jin ZHONG (Pasteur Shanghai – Chinese academy of Sciences): « Hepatitis C virus cell culture model – so much we can learn from it »
Hepatitis C virus (HCV), an enveloped positive-sense single-stranded RNA virus, can cause chronic and end-stage liver diseases. Approximately 170 million people worldwide are infected with HCV. The new annual cases of HCV infection in China have increased over the past decade and remain high. The tremendous progress has been achieved in the therapeutics of chronic hepatitis C thanks to the development of direct antiviral agents (DAAs), but the worldwide application of these highly effective DAAs is limited due to their high treatment cost. In addition, drug-resistance mutations present a potential problem as DAAs are becoming a standard therapy for chronic hepatitis C. Unfortunately, no vaccine is available for preventing new HCV infection despite many years of research. Therefore, HCV infection will still impose a big threat to human public health for many more years. The research progress in HCV field was greatly facilitated by development of cell culture model for HCV infection (denoted HCVcc) based on a novel genotype 2a cDNA clone of JFH-1 in 2005. Since then, a few other new infectious HCV cDNA clones have been established. In this presentation, I will review the progress of HCV cell culture model and discuss its remaining challenges. Particularly, I will introduce our own recent research work on developing new HCV cell culture model and the application in basic and translational HCV research.
